dc.description.abstract |
Prostate cancer is the second most common cancer in man. In Pakistan annual mortality rate of
prostate cancer is estimated at 0.9/100,000 people and rising rapidly. Many molecular
mechanisms involved in onset and progression of prostate cancer still remains unclear. The aim
of the present study is to investigate the possible cross talk between TPD52, KLF13,PKCɛ and
miRNA-223 and to find out the three dimensional structure of KLF13. 50 blood samples of
prostate cancer patients were collected from combined military hospital Rawalpindi (CMH) and
expression analysis was performed using Real Time PCR. The Fold change was calculated
through delta cycle threshold method. The statistical significance was calculated through one
way and two ways ANOVA. The expression of TPD52 is highly elevated i.e. 22 fold increased
as compared to healthy control while expression of miR-223, PKCɛ and KLF13 is lower i.e.
0.0168 fold decreased as compared to healthy controls. Expression of TPD52, KLF13, PKCɛ
and miR-223 was also analyzed according to stage, grade, and metastasis and treated as well as
non- treated patients. In the end we also find out the possible cross talk between all genes and it
was found that all genes are linked with kras and AKT pathway. Further study will help in the
development of new diagnostic marker and will provide new insights into the potential
mechanisms of oncogenesiss and metastasis of Prostate cancer. |
